Adenosylcobalamin

Adenosylcobalamin suppliers and sourcing guide

Adenosylcobalamin (cobamamide, 5'-deoxyadenosylcobalamin) is a form of vitamin B12 with a 5'-deoxyadenosyl group bound to the central cobalt atom. It is made either by recovering it from bacterial fermentation without converting it to cyanocobalamin, or semi-synthetically from other cobalamins, then purified and crystallized. It is a dark red powder that is very sensitive to light, so it is handled under subdued light and sold as pure material and as diluted powders, often blended with methylcobalamin.

Forms and grades of adenosylcobalamin

Suppliers sell different products under the same name. Specify the one you need.

  • Pure material

High-purity adenosylcobalamin for premix makers and manufacturers who dilute it themselves.

  • 1% trituration

Adenosylcobalamin on a carrier such as dicalcium phosphate or microcrystalline cellulose at 1%.

  • Blends with methylcobalamin

Pre-mixed cobalamin blends from some suppliers; check the stated amount of each form.

What to check on the COA

Compare each result with your own specification when the lot arrives.

Assay (HPLC)
Cobalamin content by HPLC (or UV for cyanocobalamin), on a dried basis for pure material and as a percentage for dilutions.
Related cobalamins
Other cobalamins and degradation products by HPLC, especially hydroxocobalamin, which forms when light-sensitive cobalamins break down.
Identity
UV-visible spectrum and HPLC retention time against a reference standard.
Loss on drying or water
Moisture, which is high in crystalline cobalamins and must be accounted for in potency.
Content uniformity (dilutions)
Assay of several samples per lot for 0.1% and 1% dilutions, to confirm even distribution.
Heavy metals and microbiology
Lead and other elemental impurities by ICP-MS, plus total aerobic count, yeast and mold and pathogens.

Storage, lots and expiry

Storage and shelf life

Adenosylcobalamin breaks down rapidly in light, so keep it in light-proof, tightly sealed containers in a cool, dry place and handle it under subdued light. Suppliers set shelf life or a retest date, with HPLC assay and related cobalamins as the usual retest checks.

Lots and expiry dates

Record assay and hydroxocobalamin content for each lot, and dose from the actual assay. Keep pure material, dilutions and blends as separate items.

Regulatory status

Facts from regulators and standards bodies, each linked to its source.

This is general information, not regulatory advice. Status differs by country and by how the ingredient is used, so check the rules for your product and market.

Frequently asked questions

What forms of adenosylcobalamin can brands buy?

Common forms include pure material, 1% trituration and blends with methylcobalamin. Ask each supplier for the specification sheet of the exact grade they quote.

What should the COA for adenosylcobalamin include?

Expect assay (HPLC), related cobalamins, identity, loss on drying or water, content uniformity (dilutions) and heavy metals and microbiology, each with its method, specification and result for that lot.

How should adenosylcobalamin be stored?

Adenosylcobalamin breaks down rapidly in light, so keep it in light-proof, tightly sealed containers in a cool, dry place and handle it under subdued light. Suppliers set shelf life or a retest date, with HPLC assay and related cobalamins as the usual retest checks.

What is the regulatory status of adenosylcobalamin?

Health Canada's Multi-Vitamin/Mineral Supplements monograph lists cobamamide as a source ingredient for vitamin B12. 21 CFR 101.9(c)(8)(iv) sets the Reference Daily Intake for vitamin B12 at 2.4 mcg for adults and children 4 years and older.
