Spermidine

Spermidine suppliers and sourcing guide

Spermidine is a polyamine found in many foods, with higher levels in wheat germ, soybeans, mushrooms and aged cheese. Supplement ingredients are usually wheat germ extracts standardized to a spermidine content, or extracts from rice germ, buckwheat sprouts or yeast; synthetic spermidine trihydrochloride is also sold. Extracts are typically low in spermidine by weight, so they are dosed by the extract amount needed to reach a stated spermidine level.

Forms and grades of spermidine

Suppliers sell different products under the same name. Specify the one you need.

  • Wheat germ extract

Extract standardized to spermidine; contains wheat, so check gluten content and allergen labeling.

  • Rice germ or buckwheat seedling extracts

Non-wheat sources, specified by spermidine content.

  • Synthetic spermidine trihydrochloride

Pure salt form; the spermidine content is lower than the salt weight.

  • Yeast-derived spermidine

Spermidine-rich yeast or yeast extract, offered by some suppliers.

What to check on the COA

Compare each result with your own specification when the lot arrives.

Spermidine assay (HPLC)
Spermidine by HPLC after derivatization, often with spermine and putrescine reported alongside.
Gluten and allergens
Gluten by ELISA for wheat germ extracts, with an allergen statement from the supplier.
Moisture
Loss on drying against the specification; extracts can be hygroscopic.
Residual solvents
Extraction solvents such as ethanol, against USP <467> or the buyer's limits.
Heavy metals
Lead, arsenic, cadmium and mercury by ICP-MS, checked against the supplier specification and the buyer's own limits.
Microbiology
Total plate count, yeast and mold, coliforms or E. coli, and Salmonella, against the supplier specification.

Storage, lots and expiry

Storage and shelf life

Store sealed in a cool, dry place; extracts can pick up moisture and cake. Spermidine salts are hygroscopic and should be kept tightly closed. Shelf life and retest date come from the supplier's specification, with spermidine assay repeated at retest.

Lots and expiry dates

Record spermidine content and source material (wheat germ, rice, buckwheat, yeast or synthetic) for each lot, since the amount of extract per serving depends on it. Keep allergen statements with the lot file.

Regulatory status

Facts from regulators and standards bodies, each linked to its source.

  • FDA's GRAS Notice Inventory lists GRN 889 for spermidine-rich wheat germ extract, notified by TLL The Longevity Labs; at the notifier's request, FDA ceased to evaluate the notice on March 20, 2020.

  • Health Canada's Natural Health Products Ingredients Database lists Chlorella vulgaris, rice seed germ and wheat seed germ as source materials for spermidine, and states it cannot be used in natural health products without additional evidence for safety.

  • The EU Novel Food Catalogue records an Austrian conclusion that flour from buckwheat seedlings with a high spermidine content is a novel food.

Frequently asked questions

What forms of spermidine can brands buy?

Common forms include wheat germ extract, rice germ or buckwheat seedling extracts, synthetic spermidine trihydrochloride and yeast-derived spermidine. Ask each supplier for the specification sheet of the exact grade they quote.

What should the COA for spermidine include?

Expect spermidine assay (HPLC), gluten and allergens, moisture, residual solvents, heavy metals and microbiology, each with its method, specification and result for that lot.

How should spermidine be stored?

Store sealed in a cool, dry place; extracts can pick up moisture and cake. Spermidine salts are hygroscopic and should be kept tightly closed. Shelf life and retest date come from the supplier's specification, with spermidine assay repeated at retest.

What is the regulatory status of spermidine?
